Dear technical kala-azar adviser, MSF-Holland,
Our experience with ELISA K39 in Study of Leishmania Reservoirs In Nuba
Mountains, western Sudan. (1999-2000).
8/1 positive. DAT- blood sera/ ELISA K39 tests in rabbits samples (total
n=14), as well
3/3 were negatives.
5/1 positive. DAT-spleen smears/ELISA K39 tests in rabbits samples (total
n=14), as well 6/6 were negatives.
One dog was tested by K39, was found to be positive.
ELISA Dip Stick Test (DST):
ELISA-DST is a modified ELISA test and performed with blood specimens (from
suspected reservoir) spotted on filter paper. Samples were cut (circle of
0.6mm) and kept in 0.9% normal saline (Nacl) overnight. In the next day the
strip was immersed in the DST-specific solution, after five minutes the
result was read, if one line was present then it is negative and if two
lines were present then the sample will be considered as positive.
This test was used in an epidemiological study to screen villagers in an
area between two hyperendemic foci, visceral and cutaneous leishmaniasis
(east and west Sudan). The final results was negative in all samples, in
fact the PCR results of 32 cases were positive and more than 50% of cases
were positive by LST. Other tools such as DAT, ELISA showed variable
results. (please have a look to our references).
